ANNEXIN V- FITC/PI 凋亡檢測試劑盒.
產(chǎn)品說明：
細(xì)胞凋亡是細(xì)胞的基本特征之一，在機(jī)體的胚胎發(fā)育、組織修復(fù)、內(nèi)環(huán)境的穩(wěn)定等方面都起到 十分重要的作用。Annexin V 是一種分子量為 35.8 KD 的 Ca2+依賴性磷脂結(jié)合蛋白，能與細(xì)胞凋亡 過程中翻轉(zhuǎn)到膜外的磷脂酰絲氨酸（Phosphatidylserine, PS）高親和力特異結(jié)合。FITC-Annexin V 結(jié) 合到凋亡細(xì)胞后，在藍(lán)色光的激發(fā)下，發(fā)出綠色熒光，區(qū)分出凋亡細(xì)胞與正常細(xì)胞。碘化丙啶 （Propidium iodide，PI）是一種核酸染料，它不能穿透完整細(xì)胞膜，但對凋亡晚期細(xì)胞和死細(xì)胞的 破損細(xì)胞膜能夠穿透，并使細(xì)胞核紅染。將 FITC-Annexin V 與PI 匹配使用，可以將凋亡早期的細(xì) 胞和晚期的細(xì)胞區(qū)分開來。 :
操作步驟
1、 用 27ml 的去離子水稀釋 3ml Binding Buffer（10×）至 30ml，每次用 3ml。
2、 收集細(xì)胞（1×106 個(gè)/次），然后用冷的 PBS 洗滌。（對于貼壁細(xì)胞，先用胰酶消化，再用 PBS 洗 滌）。
3、 用 1ml 1×的 Binding Buffer 懸浮細(xì)胞，300×g 離心 10min，棄上清。
4、 用 1ml 1×的 Binding Buffer 重懸細(xì)胞，使細(xì)胞的密度達(dá)到 1×106 個(gè)/ml。
5、 每管加入 100μL 細(xì)胞（1×105 個(gè)）。
6、 再向管中加入 5μL Annexin V-FITC。
7、 室溫，避光，輕輕地混勻，10min。
8、 加入 5μL PI，室溫，避光，孵育 5min。
9、 加入 PBS 至 500μL，輕輕混勻。
10、在 1 小時(shí)內(nèi)用流式細(xì)胞儀檢測。
注意事項(xiàng)
1、旋帽離心管裝的試劑在開蓋前請短暫離心，將蓋內(nèi)壁上的液體甩至管底，避免開蓋時(shí)液體灑落。
2、Annexin V-FITC 和 Propidium iodide 是光敏物質(zhì)，在保存與操作時(shí)請注意避光。
3、Propidium iodide（PI）能通過皮膚吸收，對眼睛有刺激作用。
4、在細(xì)胞洗滌的*一步，請盡量將上清棄凈，以免 PBS 殘留影響實(shí)驗(yàn)結(jié)果。 產(chǎn)品內(nèi)容: CA1020-20 CA1020-50 CA1020-100 保存 Annexin V-FITC 100μl 250μl 500μl 4°C，避光，不要冷凍 Propidium iodide(PI) 100μl 250μl 500μl 4°C，避光，不要冷凍 Binding Buffer（10×） 6ml 15ml 30ml 4°C 保存，長時(shí)間-20°C
5、PI 染色時(shí)間過長有可能造成檢測的凋亡率偏高，建議首先進(jìn)行 Annexin V-FITC 染色，上機(jī)前 5 分鐘再加入PI 染色。
6、整個(gè)操作過程動(dòng)作要盡量輕柔，勿用力吹打細(xì)胞，盡量在 4℃下操作。
7、反應(yīng)完畢后請盡快檢測，因?yàn)榧?xì)胞凋亡是一個(gè)動(dòng)態(tài)的過程，反應(yīng) 1 小時(shí)后熒光強(qiáng)度就開始衰變。
8、成功的檢測凋亡受以下幾種因素的影響，如細(xì)胞類型、細(xì)胞膜上 PS 的密度、發(fā)生凋亡時(shí) PS 翻 轉(zhuǎn)的比例、誘導(dǎo)細(xì)胞凋亡的方法、所用試劑、誘導(dǎo)凋亡的時(shí)間等，把這些影響因素進(jìn)行優(yōu)化對實(shí)驗(yàn) 成功是非常必要的。 此試劑盒僅供科研使用。
