產(chǎn)品簡介：
堿性條件下，蛋白將Cu2+還原為Cu+， Cu+與BCA試劑形成紫藍色的絡(luò)合物，測定其在562nm處的吸收值，并與標準曲線對比，即可計算待測蛋白的濃度。常用濃度的去垢劑SDS，Triton X-100，Tween不影響檢測結(jié)果，但受螯合劑(EDTA, EGTA)、還原劑（DTT，巰基乙醇）和脂類的影響。實驗中，若發(fā)現(xiàn)樣品稀釋液或裂解液本身背景值較高，可試用Bradford蛋白濃度測定試劑盒。
操作說明：
一. 微孔酶標儀法
1. 配制工作液：根據(jù)標準品和樣品數(shù)量，按50體積BCA試劑加1體積Cu試劑（50:1）配制成BCA工作液，充分混勻(混合時可能會有渾濁，但混勻后就會消失)。BCA工作液室溫24小時內(nèi)穩(wěn)定。
2. 稀釋標準品：取10μL BSA標準品用PBS稀釋至100μL（樣品一般可用PBS稀釋），使終濃度為0.5mg/mL。將標準品按0, 2, 4, 6, 8, 12, 16, 20μL加到96孔板的蛋白標準品孔中，加PBS補足至20μL。
3. 將樣品作適當稀釋（ 多做幾個梯度，如作2倍、4倍、8倍稀釋），加20μL到96孔板的樣品孔中。由于移液器在取小量樣品時誤差偏大，標準線前面的點可能不很準確，所以盡可能的讓樣品點落在標準線1/2后。
4. 各孔加入200μL BCA工作液，37℃放置15-30分鐘。用酶標儀測定A562nm，根據(jù)標準曲線計算出蛋白濃度。使用溫箱孵育時，應(yīng)注意防止因水分蒸發(fā)影響檢測結(jié)果。
二. 分光光度計法 如沒有酶標儀，可用分光光度計在離心管中混勻后加入比色皿中比色。 步驟如下：
1. 配制工作液: 根據(jù)標準品和樣品數(shù)量，按50體積BCA試劑加1體積Cu試劑（50:1）配制成BCA工作液，充分混勻(混合時可能會有渾濁，但混勻后就會消失)。BCA工作液室溫24小時內(nèi)穩(wěn)定。
2. 稀釋標準品：取100μL BSA標準品用PBS稀釋至1mL（樣品一般可用PBS稀釋），使終濃度為0.5mg/mL。
3. 取八支（或者更多）5mL離心管，標上號，按下表加入試劑。
4.37℃放置15-30分鐘。用分光光度計測562nm處吸光值，根據(jù)標準曲線計算出蛋白濃度。
注意事項：
1. 長期不用時，Cu試劑與PBS稀釋液可置于2-8℃保存，如發(fā)現(xiàn)細菌污染則應(yīng)丟棄。BCA試劑在低溫條件下出現(xiàn)結(jié)晶沉淀時，可37℃溫育使其完全溶解, 不影響使用。
2. 樣品中若含有較多干擾物質(zhì)時，請采用Bradford蛋白濃度測定試劑盒。
